Study suggests ‘sleeping sickness’ parasite mutated to evade immune system defences

Research teams from the universities of Georgia and Glasgow discovered the mechanism the Trypanosome parasite evolved for “sleeping sickness” disease to circumvent the human immune system. Their study follows a recent African outbreak of the disease this parasite causes. The discovery is expected to help develop a cure preventing future deaths.

The 1998–2001 sleeping sickness epidemics in South Sudan, Angola, Democratic Republic of Congo, and Uganda caused tens of thousands of deaths. The parasites are spread by tsetse flies. It is considered one of the worst epidemics in the last five decades. As WHO reported, subsequent recent introduction of population screening in rural areas and distribution of more effective drugs had potentially reduced the number of death incidents. However, the main effect of these measures being a decrease in spreading of the parasite and not in decrease of the death rate, the researchers decided to tackle the puzzle.

The scientists studied the molecules the human immune system activates in response to the attack. They were looking for the way the parasites inhibit it and lead to the death of the victim. Of the several known parasite species, the two that can infect humans were chosen to study, called Trypanosoma brucei gambiense and Trypanosoma brucei rhodensiense. Several others also exist, but they affect only animals.

Researchers knew that the Trypanosoma attacks typically are repelled by an HDL, more specifically trypanosome lytic factor-1 (TLF-1). It is generated in the humans’ immune system and is toxic to the parasites but not to humans. It was known that one of the parasites, T. b. rhodensiense, has evolved an inhibitor of the toxic chemical, called Serum Resistance Associated (SRA) protein. However the other one, T. b. gambiense, was responsible for over 95% of human deaths, and it had been previously unknown why.

The study showed that a gene mutation has lead to a change in the parasite’s surface protein receptor. It doesn’t bind to TLF-1 as well as for other species, leading to a substantial decrease in TLF-1 uptake by the parasite. The receptor is more strict, causing a lower nutrient intake as well, but it suffices, and the parasite defends against the human immune system.

As professor and head of the department of biochemistry and molecular biology at UGA (and one of the leaders of the research) Stephen Hajduk explained, “Humans have evolved TLF-1 as a highly specific toxin against African trypanosomes by tricking the parasite into taking up this HDL because it resembles a nutrient the parasite needs for survival. But T. b. gambiense has evolved a counter measure to these human ‘Trojan horses’ simply by barring the door and not allowing TLF-1 to enter the cell, effectively blocking human innate immunity and leading to infection and ultimately disease.”

Based on the results of the research, humans could possibly develop a defense mechanism to keep the parasites from evading the human defense system. Stephen Hajduk commented, “We believe this research represents a paradigm shift and causes us to think more broadly about how pathogens avoid host defense mechanisms. It turns out that African trypanosomes have evolved a diversity of ways to avoid human innate and acquired immune systems.”

Fiery Egyptian tourist bus crash kills nine

Nine people have died in a fiery tourist bus crash in Egypt on Thursday. The bus was carrying 40 North American and European passengers when it crashed in the Sinai peninsula.

The bus left the road when it skidded off a tight bend and struck a concrete barrier, rolling over and catching fire. The bus ended the roll back the right way up and was completely destroyed by the fire. One wheel lay twelve metres from the main wreckage and skid marks were left on the road. The coach had been travelling from a resort at nearby Sharm el-Sheikh to the capital Cairo when it crashed at Abu Zenima, with some reports suggesting the cause was a blowout on a tyre.

According to the Egyptian security official eight Russian nationals were killed, as was one of two drivers on board, both of whom were Egyptian. This was contradicted by Russian state owned service RIA Novosti, which stated just two Russians were among the dead. The Russian foreign ministry only said 16 Russians were injured and none killed, also contradicting the Egyptian security official who said only 14 people were injured in total. The Ukrainian foreign ministry said a Ukrainian woman had been killed and the Romanian foreign ministry said a 41-year-old Romanian woman was amongst the dead. The driver behind the wheel at the time survived the accident.

The crash occurred at 6:00 am (0300 GMT) about 70km (45 miles) from Suez, and many of the injured were rushed to hospital there. Traffic officials, security personnel, civil defence soldiers and ambulances all responded to the crash. In addition, the Azure Travel coach, like all Egyptian tour buses, had at least one armed police officer travelling on board.

Other injured passengers were taken to Sharm. Two Canadians and two British citizens are reported to be amongst the injured. The Brits are in a serious condition and one Canadian woman had her hand amputated. At least one person died in hospital. It is also reported that Italians were injured in the crash.

Road accidents claim around 6,000 lives and injure 30,000 every year in Egypt. This high accident rate is fed by poor maintenance and regulatory enforcement and reckless driving, with many roads allowing high rates of speed to be achieved. A fog-triggered pile-up killed 29 in February, and 23 died in March in a head-on collision between two lorries.

Atlantis launched, heads for International Space Station

In the first space flight of 2007, Space Shuttle Atlantis blasted off on schedule yesterday, starting on an 11-day mission to the International Space Station.

Atlantis and its seven astronauts lifted off at 7:38 p.m. EDT (2338 GMT) from the launch pad at Kennedy Space Center, Florida. The launch pad, newly refurbished by NASA, had not been used since the launch of Space Shuttle Columbia, which ended in disaster on Feb. 1, 2003.Atlantis is scheduled to dock with the space station at around 1930 GMT tomorrow.

The goal for the Atlantis crew is to carry out the two missions that were left incomplete from the previous flight due to broken and jammed equipments. During the flight, the shuttle and its seven astronauts will deliver a new segment and will install energy-producing solar panels to the International Space Station. The payload is the heaviest ever flown to the space station, and includes a 45-foot (14-meter), 35,678-pound (16,183 kilogram) aluminium segment that will become part of the station’s superstructure.

The mission, STS-117 has been dogged by trouble. It was scheduled to launch in March, but a hailstorm in February damaged the orange external fuel tank, so the shuttle had to be rolled back into the Vehicle Assembly Building for repairs. After repairs, the tank had white specks visible in the orange insulation, and there was some concern over whether the insulation would hold. Falling foam insulation was blamed for the Columbia crash, in which all seven astronauts aboard were killed.

But yesterday’s launch appeared to be flawless. “Give me more speckled tanks,” the shuttle program manager, N. Wayne Hale Jr., was quoted as saying.

The delay in the launch of Atlantis also pushed back other missions.

NASA faced a scandal earlier in the year when one of its astronauts, Lisa Nowak, was fired after being charged in the attempted kidnapping of a fellow astronaut, William Oefelein, with whom she had been having an affair.

Google launches Google Spreadsheets

Google has launched an online spreadsheet site, in a private beta.

The site will allow spreadsheets to be shared between up to 10 users, which is aimed to be useful to teams and small businesses. “Many people already organise information into spreadsheets. Where they are struggling is to share it” said the product manager, Jonathan Rochelle.

Google recently bought the online word-processor Writely, launched a calendar product, as well as a desktop search tool. Many see this as them straying into Microsoft‘s markets.

Google Spreadsheets uses very advanced AJAX (Asynchronous Javascript And Xml) and Client Side Scripting to mimic very effectively it’s desktop counterparts’ functions.

It is surprisingly fast, has very good formatting and advanced formula support, but best of all it has complete support for Microsoft Excel .xls files, and very good collaboration: just enter the e-mail address and you can share easily.

Microsoft claims 235 patent breaches by open source software

Software giant Microsoft’s chief lawyer Brad Smith claimed in an interview published in the magazine Fortune on Monday that open-source software products violate 235 of Microsoft’s patents. The main transgressors are claimed to be Linux (107 patents) and OpenOffice.org (45), with e-mail programs infringing 15 patents. Microsoft wants royalties to compensate for the patent breaches.

According to Microsoft’s Vice-President of intellectual property and licensing, Horacio Gutierrez, the company wants to negotiate with the open-source companies rather than sue them. “If we wanted to litigate we would have done that a long time ago. Litigation is not an effective way of going about solutions,” Gutierrez said. According to him, Microsoft has over the last years tried to work towards a “constructive” solution to the alleged problem of patent violation.

Microsoft in the past has used the strategy of cross-licensing to get royalties from companies who infringe their patents, for example in their deal with Novell. On a company blog, Novell reiterated that their deal “is in no way an acknowledgment that Linux infringes upon any Microsoft intellectual property.”

“We don’t think that customers will want to continue on without a solution to the problem,” Gutierrez said about Microsoft’s approach to guaranteeing companies that they won’t get sued because they use the allegedly patent-infringing Linux operating system.

The upcoming third version of the GPL licence, the licence under which Linux is released, will prohibit Linux distributors to agree to patent royalty deals. Microsoft called these “attempts to tear down the bridge between proprietary and open-source software that Microsoft has worked to build with the industry and customers.”

A related U.S. Supreme Court ruling from April 30th showed how software patents can be subject to court challenges; basically, if the innovations patented are “obvious”, the patent is weakened. Joe Lindsay, information officer for a mortgage company, pointed out that the Unix code that Linux is based upon preceded Microsoft Windows, which might also be a reason for some patents to be invalid.

Red Hat, the biggest Linux distributor, said in a statement on Monday:

The reality is that the community development approach of free and open source code represents a healthy development paradigm, which, when viewed from the perspective of pending lawsuits related to intellectual property, is at least as safe as proprietary software.
 

Larry Augustin, former CEO of a company called VA Linux (now VA Software), responsible among other things for launching SourceForge.net, an open-source software development community, posted a message on his blog under the title “It’s Time for Microsoft to Put Up or Shut Up”:

If Microsoft believes that Free and Open Source Software violates any of their patents, let them put those patents forward now, in the light of day, where we can all evaluate them on their merits. If not, then stop trying to bully customers into paying royalties to use Open Source.

According to the Fortune report, more than half of the Fortune 500 companies are estimated to use Linux in their data centers.
